visible single stranded DNA
Chemistry 2009. 6. 11. 07:15 |This is single stranded DNA chemically synthesized by AKTA oligopilot 100plus (GE Healthcare).
After the solid phase synthesis with phosphoramidites, crude mixture was purified by HPLC and then desalted. Lyophilization of the solution resulted white ribbon-like phorous oligonucleotide shown in picture (the shown tube is blue capped 50ml falcon-tube). It is hard to obtain pure solid DNA with known sequence in visible amount by eyes.
순수한 DNA는 무슨 색일까? 시판하는 실험용 DNA는 순백색이다. 주로 연어의 정액이나 송아지의 분비기관에서 추출한 DNA를 쉽게 비교적 싼(?) 가격으로 얻을 수 있는데, 그 염기쌍(보통의 경우 이중나선구조, double-stranded)의 대략적인 숫자만 알 뿐이지 염기 배열(sequence)은 알지 못한다.
알고있는 정확한 sequence 의 DNA를 얻기 위해서는 화학적으로 합성하거나 분자생물학적 방법을 통해 E. coli. 등의 유전자(gene)를 알고 있는 박테리아에서 분리해 낼 수 있다.
